Abstract
Frusemide is a loop diuretic used for hypertension, heart failure among many other indications. It has an absorption window in the stomach and it's a class IV drug according to BCS with low bioavailability.
Floating tablets are one of the dosage forms that is best suited for drugs with such absorption window and gas generating excipients can be used to reduce floating lag times even more than traditional floating tablets that relay on density difference alone.
The aim of the study was to investigate the feasibility of floating tablets as a convenient dosage form for oral administration of frusemide aiming to improve its bioavailability through extended residence time in the stomach.
Eighteen formulas were prepared and pressed into tablets with manual compress and the resultant tablets were subjected to floating lag and buoyancy tests and the five best formulas (F4, F7, F9, F15 and F17) were forwarded for further testing of friability, weight variation, disintegration and dissolution testing.
These five formulas all passed weight variation and friability test successfully, had very fast disintegration times (probably faster than intended) and dissolution profiles that didn't go very well with intended aim of the study to prolong their residence time in the stomach while offering extended time for absorption in the stomach. These formulas released around 80% of their drug load within the first hour and around 100% within 8hours.
